    Gastrointestinal helminths are ubiquitous in horse populations across the world. Intensive anthelmintic treatments have succeeded in controlling some of the pathogenic effects of these parasites. However, the success of anthelmintic drugs has been threatened by the development of widespread resistance to those most commonly used. To develop improved control strategies, information on helminth distribution patterns is needed, which can be obtained by identifying animals regarded as high egg shedders and taking age, gender, and other risk factors into account. The aim of this study was to determine the prevalence of helminth infection in the Irish equine population using faecal egg counts and to evaluate the effects of risk factors on these faecal egg counts. For the 2700 horses that were included in the study, the prevalence of gastrointestinal helminth infection was 52.40%, 4.22%, 2.59%, and 0.89% for strongyle species, Parascaris spp., Anoplocephala spp., and Strongyloides westeri, respectively. Overall, strongyle faecal egg counts from 159 farms averaged 250.22 eggs per gram. Both age and season had significant effects on strongyle egg shedding. In conclusion, this study revealed high prevalence of strongyle worm infection in horses on Irish farms, which highlights the need to optimize and develop good management practices and strategic deworming.

    BACKGROUND: To identify any universal impact of strongyle parasite burden on the growth rate of young cattle.
    METHODS: A meta-analysis and meta-regression of the relationship between differences in strongyle parasite burden between cohorts and average daily weight gain was conducted. Publications were identified from a search of databases applying PRISMA 2020 principles. Eligible studies had at least two groups of growing cattle on the same farm that were identical in composition, management and diet except for parasite exposure and were subject to body weight gain or average daily gain and faecal egg count measurements across the common growing period. The reference group had the lowest growth-period faecal egg count. A meta-regression estimated the impact of strongyle parasitism. The dependent variable was the log of the ratio of average daily gain between comparison groups and the reference group with the predictor variable as the common logarithm of the difference in average faecal egg count (plus 1) between the comparison and the reference groups.
    RESULTS: 26 publications containing 85 groups and 59 comparison ratios were analysed. Papers included representatives from dairy and beef industries and from pasture and feedlot production systems and from all cattle-producing continents. The comparison group average daily growth rate was 0.89 (95%CI 0.81-0.97) that of the reference group. Meta-regression identified a 0.131 linear reduction in average daily weight gain ratio for every log10 increase in the difference between comparison and reference group faecal egg count. Direction of effect was consistent across all subset analyses (continent, class of stock and production system). Whilst small faecal egg count differences between the comparison and reference groups often provided similar rates of daily weight gain, the trend was negative with most comparison groups having lower daily weight gains than their reference group.
    CONCLUSIONS: Strongyle parasitism of growing cattle as measured by faecal egg count is associated with reduced growth across all production systems, geographies and classes of cattle.

    Global livestock production is facing serious new challenges, including climate-driven changes in parasite epidemiology, and anthelmintic resistance, driving a need for non-chemotherapeutic methods of parasite control. Selecting for genetic resistance to gastrointestinal nematode infection could reduce reliance on chemical intervention and mitigate increases in parasite challenge due to climate change. Ewes of the composite Exlana breed with a range of estimated breeding values (EBVs) based on nematode faecal egg counts (FECs) were monitored during the peri-parturient period on two farms in southwestern England. Ewes with low EBVs (\"resistant\") had lower FECs during the peri-parturient period than those with high EBVs (\"susceptible\"): the mean FEC was reduced by 23% and 34% on Farms 1 and 2, respectively, while the peak FEC was reduced by 30% and 37%, respectively. Neither EBV nor FEC were correlated with key performance indicators (estimated milk yield, measured indirectly using 8 week lamb weight, and ewe weight loss during lactation). Simulations predict that the reduced FECs of resistant ewes would result in a comparable reduction in infection pressure (arising from eggs shed by ewes) for their lambs. Furthermore, although the reduced FECs observed were modest, simulations predicted that selecting for nematode resistance in ewes could largely offset predicted future climate-driven increases in pasture infectivity arising from eggs contributed by these ewes. Selective breeding of the maternal line for nematode resistance therefore has potential epidemiological benefits by reducing pasture infectivity early in the grazing season and alleviating the need for anthelmintic treatment of ewes during the peri-parturient period, thus reducing selection pressure for anthelmintic resistance. These benefits are magnified under predicted future climate change. The maternal line warrants more attention in selective breeding programmes for nematode resistance.

    Faecal egg counts (FEC) are used widely for monitoring of parasite infection in animals, treatment decision-making and estimation of anthelmintic efficacy. When a single count or sample mean is used as a point estimate of the expectation of the egg distribution over some time interval, the variability in the egg density is not accounted for. Although variability, including quantifying sources, of egg count data has been described, the spatiotemporal distribution of nematode eggs in faeces is not well understood. We believe that statistical inference about the mean egg count for treatment decision-making has not been used previously. The aim of this study was to examine the density of Parascaris eggs in solution and faeces and to describe the use of hypothesis testing for decision-making. Faeces from two foals with Parascaris burdens were mixed with magnesium sulphate solution and 30 McMaster chambers were examined to determine the egg distribution in a well-mixed solution. To examine the distribution of eggs in faeces from an individual animal, three faecal piles from a foal with a known Parascaris burden were obtained, from which 81 counts were performed. A single faecal sample was also collected daily from 20 foals on three consecutive days and a FEC was performed on three separate portions of each sample. As appropriate, Poisson or negative binomial confidence intervals for the distribution mean were calculated. Parascaris eggs in a well-mixed solution conformed to a homogeneous Poisson process, while the egg density in faeces was not homogeneous, but aggregated. This study provides an extension from homogeneous to inhomogeneous Poisson processes, leading to an understanding of why Poisson and negative binomial distributions correspondingly provide a good fit for egg count data. The application of one-sided hypothesis tests for decision-making is presented.

    Research has shown that Toll-like receptor 4 (TLR4) is important in immune responses to some helminth parasites. In sheep, variation in the PAMP region of TLR4 may result in structurally and thus functionally different TLR4 molecules, and this may consequently lead to variation in the TLR4 response to parasite infections. This study involved three separate, but related sheep breeds (Merino, Polwarth and Corriedale sheep) and a total of 885 lambs from five New Zealand farms that underwent a mixed field-challenge from gastro-intestinal parasites. Faecal samples were collected at approximately 4 and 9 months of age and faecal egg counts (FECs) for Nematodirus spp. and Strongyle species determined, along with the total number of eggs per gram (EPG). Analysis of the five farms collectively revealed an association (P=0.023) between the presence of TLR4 variant *02 (mean 24 EPG) and the absence of the variant (mean 32 EPG) at 9 months of age. Conversely the presence of *03 had a significantly (P=0.047) higher mean Nematodirus spp. FEC (mean 42 EPG) compared to the absence (mean 28 EPG) at 9 months of age. More associations were revealed when the data were split according to the dominant faecal parasite species. With a predominantly Trichostrongylus spp. FEC group of lambs at 9 months of age, the presence of TLR4 variant *02 was found to have significantly (P=0.003) lower Nematodirus spp. FEC (mean 4 EPG), and also significantly (P=0.033) lower total FEC (mean 312 EPG) when compared to sheep without the variant (mean 15 EPG and 449 EPG, respectively). The presence of TLR4 variant *03 and *04 were associated or tended to be associated (P=0.010 and P=0.088, respectively) with higher Nematodirus spp. FEC (mean 25 EPG and 22 EPG, respectively) when compared to lambs without the variant (mean 10 EPG and 11 EPG, respectively). These results suggest that TLR4 variation may be affecting the immune response to gastro-intestinal parasites in sheep, although principally to Nematodirus spp. infections and not Strongyle species infections.
