Growing work demonstrates the ability of EVs to facilitate long-distance and cross-kingdom communication. Their innate barrier crossing and cell targeting properties make them a uniquely useful starting ground for novel drug delivery platforms. To better understand the endogenous activity and therapeutic potential of EVs, recent work has measured particle circulation and distribution in vivo using several approaches. Here, we describe molecular-based methods for quantifying bacterial EV distribution in collected tissue samples for biodistribution studies. These methods are important for understanding cell-cell communication facilitated by bacterial EVs and for identifying opportunities for using bacterial EVs as a therapeutic platform.
